4About TIGM
- Originally created by the State of Texas as a
non-profit independent organization - Joint institute of Texas AM University and Texas
AM Health Science Center - Operates the worlds largest gene trap library to
create knockout mice - Provides a variety of mouse model research
services for our own knockout models as well as
mouse models from other institutions
5Our Mission
- Discover breakthroughs in science and medicine
and accelerate the pace of medical discoveries
through internal research and by providing our
resources and services to the scientific community
6Headquarters and Research LabsHouston, Texas
- Located in the Texas Medical Center
7Mouse FacilityCollege Station, Texas
- Capacity for 80,000 genetically engineered mice
- Full shower-in barrier including animal, surgery,
necropsy, procedure, and neurobehavioral
phenotyping rooms.
8TIGM Gene Trap
- Retroviral insertion of gene trapping vector
containing a promoter-less marker/reporter gene
(Neo, or ß-Geo)
9C57BL/6 Library Construction
10C57BL/6N Gene Trap Library
- Worlds largest C57BL/6 knockout mouse library
- gt350,000 ES cell clones
- gt10,000 unique genes
11Additional TIGM Resources
- Access to 129/SvEvBrd Gene Trap and Gene Targeted
Library - over 272,000 ES cell clones
- more than 9,000 unique genes
Combined total of over 640,000 clones containing
over 13,000 unique genes
12Quality Control
Quality control procedures for ES cell clones
- qPCR for Neo to ensure one copy of gene trapping
vector is present in the genome - qPCR for SRY to ensure there is no loss or
degradation of the Y chromosome - iPCR to ensure the gene trapping vector is in the
correct location and orientation - Thioglycolate and PCR assays to monitor yeast,
mold, bacteria and Mycoplasma contamination
13Quality Control
Quality control procedures for mice
- Clones undergo quality control procedures before
they are microinjected - Tail samples are genotyped with wild type and
mutant strategies specific for the insertion site
before the animals are shipped
14Online Database/Ordering
- TIGM has an extensive, user-friendly database
accessible via the Internet. It allows you to
search through our C57BL/6 or the 129SvEvBrd gene
trap libraries, inquire about a specific line,
and place an order. - You can search by
- Entrez Gene ID
- Gene symbol
- Keyword
- Sequence in FASTA format
- Chromosome

15Online Database/Ordering
TIGMs database links directly to the UCSC Genome
Browser and allows you to view the sequence tags
generated from our vector insertion sites. Click
the TIGM KNOCKOUTS link at the top of the UCSC
genome browser window to view all of TIGMs
available clones.
16TIGM Services
