Lenalidomide An Immunomodulatory Medication
  • Lenalidomide, is a medication used to treat
    multiple myeloma (MM) and myelodysplastic
    syndromes (MDS). For MM it is used after at least
    one other treatment and generally together with
    dexamethasone.
  • Side Effects Of Lenalidomide-
  • Lenalidomide sometimes causes side effects due to
    the rapid destruction of cancer cells (tumor
    lysis syndrome).

Precautions and Warning
  • Embryo-Fetal Toxicity- An embryo-fetal
    development study in monkeys indicates that
    lenalidomide produced malformations in the
    offspring of female monkeys who received the drug
    during pregnancy, similar to birth defects
    observed in humans following exposure to
    thalidomide during pregnancy.
  • Hematologic Toxicity- Patients taking
    Lenalidomide for MM should have their complete
    blood counts monitored every 2 weeks for the
    first 12 weeks and then monthly thereafter.
  • Venous Thromboembolism- Venous thromboembolic
    events (predominantly deep venous thrombosis and
    pulmonary embolism) have occurred in patients
    with multiple myeloma treated with lenalidomide
    combination therapy see Boxed Warning and
    patients with MDS or MCL treated with
    lenalidomide monotherapy.
  • Tumor Flare Reaction- Tumor flare reaction has
    occurred during investigational use of
    lenalidomide for CLL and lymphoma, and is
    characterized by tender lymph node swelling, low
    grade fever, pain and rash. Treatment of CLL with
    lenalidomide outside of a well-monitored clinical
    trial is discouraged.

Indications and Usage
  • Multiple Myeloma- Lenalidomide in combination
    with dexamethasone is indicated for the treatment
    of patients with multiple myeloma (MM) who have
    received at least one prior therapy.
  • Myelodysplastic Syndromes- Lenalidomide is
    indicated for the treatment of patients with
    transfusion-dependent anemia due to low- or
    intermediate-1-risk myelodysplastic syndromes
    (MDS) associated with a deletion 5q cytogenetic
    abnormality with or without additional
    cytogenetic abnormalities.
  • Mantle Cell Lymphoma- Lenalidomide is indicated
    for the treatment of patients with mantle cell
    lymphoma (MCL) whose disease has relapsed or
    progressed after two prior therapies, one of
    which included bortezomib.

Availability
  • The lenalidomide 10 mg is also used for anemia
    (reduced or low RBCs) in candidates with MDS
    (myelodysplastic syndrome) caused by certain
    abnormal chromosome. In addition, lenalidomide
    capsule 10 mg is also used for mantle cell
    lymphoma, follicular lymphoma, or marginal zone
    lymphoma, after other medicines have been
    considered without success.
