5Step 1
- Fix the tissue sample
- After collection, transfer the tissue into a
container of 10 neutral buffered formalin - Cap the container tightly and keep so for 24-48
hours - Ensure the container is not leaking and label it
correctly
6Step 2
- Transfer the tissue to histology cassette
- Trim the tissue specimen using a scalpel
- Make it appropriate to fit into the cassette
- Keep the edges of the specimen thin, less than
4mm - Store the cassette in formalin
7Step 3
- Process the tissue specimen
- Immerse the specimen in increasing level of
alcohol to remove any water or formalin from it - Clean the specimen with xylene
- Embed the specimen in paraffin wax
- Ensure the wax surrounds the tissue and forms a
block
8Step 4
- Section the tissue specimen
- Remove wax from tissue surface
- Keep the tissue block in a refrigerator for 10
minutes - Slice the specimen into thin ribbons using a
microtome - Transfer the tissue slices to a warm water bath

- Place a charged slide under the water level to
scoop the tissue slices on the slide surface - Label the slides correctly
- Allow them to dry in upright position at 37C
10Step 5
- Stain the slides
- Use a histochemical stain like hematoxylin or
eosin to stain the tissue specimen on the slide - Attach a cover slip after staining for long-term
protection of the slide
