Letter e Experiment
  • Objectives
  • Students will learn the function of each part of
    a microscope.
  • Students will observe that the letter "e" is
    upside down when viewed under low power.
  • Students will notice when the slide with the
    letter "e" is moved in one direction, it appears
    to move in the opposite direction.
  • Students will observe how the letter "e" looks
    when viewed from low to medium and to high power.

Wet mount slide
Onion cells
  • Remove a thin piece of onion skin tissue from the
    onion
  • Place the tissue on a slide
  • Add a drop of iodine stain
  • Carefully place the cover slip on top of the
    onion cells
  • Observe under low, medium and high magnification
  • Record observations

  • Scrape inside of cheek with a cotton swab
  • Place the tissue on a slide
  • Add a drop of methylene blue stain
  • Carefully place the cover slip on top of the
    cheek cells

Staining
  • Staining is required to view the cells under
    the microscope
  • Onion cell Iodine stain
  • Reacts with starch to stain cells brown, making
    nuclei more visible
  • Cheek cell Methylene blue stain
  • Stains cells blue, making nuclei more visible
