  • General Design Guidelines
  • 1. Find 21 nt sequences in the target mRNA that
    begin with an AA dinucleotide.
  • 2. Select 2-4 target sequences.
  • - GC content 30-50
  • - 5-6 T's(terminate transcription)
  • - avoid intron regions and stretches of 4 or
    more bases
  • - perform BLAST homology search
  • 3. Design appropriate controls.

  • - Additional siRNA sequences targeting the same
    mRNA.
