Materials

• Nutrient Broth
• Petri Dishes
• Blood Agar Plates
• Cotton Swabs
• Inoculating wire loop
• Forceps
• Sterile disk
• Sterile water
• Tape
• Wax pencil
• Bunsen burners
• Cleaners

Method

• Prepare Nutrient Agar Plates
• 1. Suspend 23g of NA powder in 1L purified H2O
• 2. Mix and heat until power is dissolved and
  solution is clear
• 3. Autoclave _at_ 121C for 15 min.
• 4. Fill as many petri dishes as you can ½ way
  with NA and let them solidify

• Collect Samples
• 1. Dip cotton swab in sterile H2O and take
  samples of skin, throat, environment, and hand.
  Rub the swab in NA plate. Cover and label.
• 2. Tape all the plates together and place them
  inverted into the incubator at 37C.

• Isolation-pure Culture
• (Discontinuous Streaking)
• 1. Sterilize wire loop by holding over flame
  until it turns bright orange
• 2. Wait a few seconds, cool loop by lightly
  touching NA
• 3. Take bacteria with loop from sample culture
  and streak on other plate.
• 4. Cover, label, and tape plates.
• 5. Put in incubator at 37C

• follow a different pattern.

• Resuspend bacteria
• 1. Pour 10ml Nutrient Broth into continuously
  streaked plates.
• 2. Swab bacteria until all is suspended in
  liquid
• 3. Swab bacteria into new plates.
• 4. Dip sterile disk in cleaners and place in
  center of plates. Tap gently to secure.
• 5. Label plates and place them inverted in the
  incubator.

• 6. Let plates sit over night.
• 7. Measure the zone of inhibition and make your
  conclusions.
