Title: Assembly and analysis of DNA

1
Assembly and analysis of DNA
2
What is a sequence assembly?
  • Putting together pieces of the DNA that was
    previously sequenced.
  • Also means align, edit and correct errors in
order to obtain a consensus sequence.

3
Some key features of successful editors
  • Use of color or symbols to illustrate key
    features (different bases, conserved regions)
  • Ability to see the traces files
  • Ability to display the complementary strand
  • Possibility to edit sequences
  • And others

4
How to calculate the average coverage of the
consensus?
  • 2 ways to do that
  • ( seq reactions X of bases)/gene or genome
    size
  • Ex 20 seq.reactions X 500bases/ 2,000 5
  • - Select several parts of the consensus sequence
(ex beginning, middle and end) and see how many
seq. reactions was needed and take the mean of
    those.
