Herbicide Resistance
  • Germination Rates
  • Spore samples from gt 10 populations
  • 100 spores (n 10) from each sample
  • Place in agar media
  • Count germinating spores

17
Spore Counts
1) Weigh out known amount of spores 0.03
grams 2) Suspend in 1 ml of water 3) Take 0.1 ul
sample and count with Haemocytometer 4) Spores
estimates - 289200 spores / 0.03 g / 1 ml water
18
Metsulfuron methyl(exposure rates)
  1. Expose spores in nutrient / agar herbicide
    solution
  2. Determine resistancerate
  3. Allow resistancebiotype to matureand collect
    spores
  4. Repeat experiment
  • 0.05 oz
  • 0.10 oz
  • 0.25 oz
  • 0.50 oz
  • 1.00 oz
  • 1.50 oz
  • 2.00 oz
  • 3.00 oz
  • 4.00 oz
  • 8.00 oz

(at 20 and 100 gal/ac)

Conclusions
  • Translocation work indicates that triclopyr
    translocates significantly better than glyphosate
    and metsulfuron.
  • Glyphosate, metsulfuron methyl, and triclopyr
provided excellent control of L. microphyllum
    under greenhouse trials.
  • In field trials, tanks mixes provided better
control than single herbicides, but results are
    preliminary.
  • Investigations on the ability of L. microphyllum
to exhibit resistance to metsulfuron methyl are
    being conducted
