Todays Job
  • Draw Hexa-Methionine (Met6 short protein)
  • Draw Met12

3
What is protein?
  • Protein is polymer of Amino Acids.

4
What is Methionine?
Side Chain
Backbone
It is one of amino acids.

Amino Acids ? Protein
7
Use Templates in ISIS Draw
  • Open ISIS Draw
  • Select Templates Amino Acids
  • Click N in Methionyl

8
Draw Met2 on ISIS Draw
  • At any empty space, click once to get a single
    methionine.
  • Now, click C in CO to get Met2, such as

9
Draw Met6 on ISIS Draw
  • Continue to click C in CO to get Met3, and then
    Met4, Met5, Met6.
  • Click Select tool
  • Select the whole molecule
  • Then, Select Edit Copy

10
3D View of Met6
  • Open WebLab
  • Select Edit Paste
  • Click Clean twice
  • Click Add Hydrogens
  • Click Clean twice
  • Please identify the position of red square

11
Copy, Paste and Move Met6
  • Select Edit Select All
  • Select Edit Paste
  • Select Translate Tool
  • Both push Ctrl and drag the mouse to move
yellow-spotted molecule up for some distance such
    as

2 Met6 ? Met12
  • Please identify N and C terminals for both two
    Met6
  • Select Sketch
  • Click a C terminal on one Met6 and a N terminal
on another Met6 to get Met12
  • Then click Clean to get helix structure of Met12

14
Keep Cleaning Structure
  • Remember the distance of the pitch
  • Keep on cleaning the helix structure of Met12
until that the pitches are not changed

Homework 6
  • 1. What is the average distance of the pitch?
  • 2. How many methionine residues are in one pitch?
  • 3. Are there any H-bonding in Met12? (Please
select Tools Monitors Hbond to investigate
    H-bond)
  • 4. Please draw 3-D structure of the following
    molecules?
