  • GPCRs belong to a very large family of proteins,
    which mainly involves in the signal transduction,
    whose topological conformation is the receptors
    of seven transmembrane. When the ligands outside
    membrane are connected with receptors, the
    interior of the receptors will mutually bind with
    each other and then activate the G-protein. While
    as for G-protein, there are two approaches to
    transmit extracellular information. The first
    approach is to open the transmembrane ion
    channels so that the ions can enter from outside.
    The second is to activate a second messenger, sch
    as cAMp and DAG. In fact, Calcium ions are
    guenerally considered as the third messengers.

  • Generally speaking, GPCRs are structurally
    flexible and dynamic because they can apply
    different structures, namely the conformation, to
    diverse factors. It is believed that a
    conformational equilibrium exists in GPCRs
    molecule between active and inactive biophysical
    states so it is necessary to pay more attention
    to the binding of ligands to GPCRs for they might
    change the equilibrium toward the active
    receptors. In terms of the current social
    reality, molecular drug targets have become
    extensively important in the application of
    modern new drugs, and drug targets might be more
    effective with the further research of GPCRs.
