Label a sterile 50 ml Conical tube with your name
and date.

Do a sterile transfer of one sterile pipette full
of LB broth containing ampicillin antibiotic
into the Conical tube.

Locate a well-formed colony on your plate of
transformed bacteria. Use a sterile inoculating
loop, bent paper clip, or toothpick to scrape up
the colony and transfer it to the LB broth in the
tube.

Overnight Culture SuspensionStep 4
Incubate the tube for 24 hours in a 37oC shaking
water bath or 48 hours in a 37oC incubator with
occasional shaking by hand (shake the tube a
couple of times an hour when possible).

After the overnight growth period is complete,
proceed to part II, Plasmid Purification.
